چکیده

Amiloride (AM), N-amidino-3,5,-diamino-6-chloropyrazinamide hydrochloride, induces natriuretic effects without potassium loss or rise in serum uric acid. 1 , 4 In single doses, AM has been shown to be less potent than hydrochlorothiazide (He); however, when administered daily in approximately maximal dosage, the natriuretic action of AM is sustained for a longer period so that the negative sodium balance equals that of He after 5 days of administration. 5 It seemed desirable therefore to investigate

Amiloride hydrochloride in hypertensive patients.

In man amiloride hydrochloride (MK-870) has been shown to be an orally effective diuretic which causes a positive potassium balance. It also enhances the saluretic effects of hydrochlorothiazide and ethacrynic acid and in addition prevents the negative potassium balance caused by these diuretics (Wilson et al., 1966; Gombos et al., 1966). Gombos et al. also studied its effects in 10 hypertensiv...

Maximising antihypertensive effects of angiotensin II receptor blockers with thiazide diuretic combination therapy: focus on irbesartan/hydrochlorothiazide

BACKGROUND Evidence-based guidelines for the management of hypertension are now well established. Studies have shown that more than 60% of patients with hypertension will require two or more drugs to achieve current treatment targets. DISCUSSION Combination therapy is recommended as first-line treatment by the JNC-7 guidelines for patients with a blood pressure > 20 mmHg above the systolic go...

Role of macula densa in diuretics-induced renin release.

Diuretic therapy may enhance renin release by various mechanisms, principally contraction of extracellular fluid volume and its effects, including a fall in arterial pressure. Awake hydropenic or volume-expanded rats received diuretics (amiloride and hydrochlorothiazide) that are known inhibitors of NaCl transport beyond the macula densa; also the well-known Na(+)-K(+)-2 Cl- transport system in...
